PickFluo

Fluorescence imaging in the absence of darkness

Category

PROFESSIONAL PHOTOGRAPHY

A revolutionary set of procedures, software, and calibration that allows you to obtain the right fluorescence even in the presence of light pollution: PickFluo is the software tool designed to guide the entire process in the absence of darkness.

PickFluo Fluorescence Imaging in the Absence of Darkness

Classically, fluorescence images are obtained from light in one band and a sensor tuned to another band of longer wavelength. The most commonly used type is UV-VIS, but UV-IR and VIS-IR types are also used. What they have in common is that the light source must not pollute the acquisition band, and this requires a dark room to do it properly. But this is not always possible.

PickFluo was created to solve this critical issue.

Phases

Acquisition

Ambient light should be reduced as much as possible. With such low light, the camera, set to the correct exposure, will need to take one or more images (more than one must be averaged to increase the signal-to-noise ratio).

Next, the excitation light must be turned on (UV, for example) without changing anything in the scene or the camera settings, and additional images must be taken.

Acquisition in PickFluo

PickFluo acquires all the images taken and processes the multiple shots to extract the pure fluorescence image, also offering an interface tool to correct the balance between the RGB channels.

Alignment

Alignment of acquired UV-Visible images.

Final result

PickFluo covers fluorescence imaging with UV and VIS excitation, and VIS and IR detection (UV-VIS, UV-IR, VIS-IR). The fluorescence images obtained can be integrated into the workflow either by reading data from SpectraPick or by reading images from PickVIewer.
